The MaxSignal® Methotrexate ELISA Kit uses a competitive immunoassay method to determine the amount of methotrexate present in a serum or urine sample. Test plate wells are coated with drug. Blood or urine sample is added for analysis, along with antibody against methotrexate. Methotrexate in the sample will compete for the primary antibody, thereby preventing the antibody from binding to the drug attached to the well. After incubation, the sample is removed, the wells are washed and a secondary HRP-conjugated antibody is added. The intensity of the absorbance at 450 nm is directly proportional to the amount of drug in the sample.
Methotrexate belongs to a group of drugs known as anti-metabolites. It acts by inhibiting the metabolism of folic acid. Methotrexate is used in treatment of cancer, autoimmune diseases and as an abortifacient in the induction of medical abortions. In racing, human recombinant Erythropoietins have been used in horse doping. It is believed that Methotrexate is also given to horses undergoing this treatment to prevent the horse from mounting an immune response to the foreign human recombinant protein.
