ELISA for Rapid Screening of Histamine Contamination
The MaxSignal® Histamine ELISA Test Kit, a colorimetric ELISA assay for the determination of histamine in fresh or frozen fish and fish meal, is the most sensitive and rapid ELISA commercially available. The plate wells are coated with anti-histamine antibody. During analysis, sample is added along with the histamine-horseradish peroxidase (Histamine-HRP) conjugate. If histamine residue is present in the sample, it will compete for the Histamine antibody, thereby preventing the histamine-HRP from binding to the antibody attached to the well. The resulting color intensity, after addition of the HRP substrate (TMB), has an inverse relationship with the concentration of histamine in the sample.

Optimized Sample Prep Protocols
This ELISA uses a rapid (15 minute) aqueous extraction method which does not require heating or organic extraction steps. Unlike other histamine ELISAs, the MaxSignal Histamine ELISA does not require time-consuming and laborious acylation steps.
Histamine is a naturally occurring contaminant found in scombroid fish such as tuna, mahi mahi, swordfish and mackerel that have been improperly handled. High levels of histamine in seafood can cause scombroid poisoning. Quality fish has histamine levels below 50 ppm. The amount of histamine can be determined using HPLC or LC-MS. However, these traditional methods require expensive instrumentation and time-consuming sample preparation procedures that can be eliminated by ELISA methodology.

The MaxSignal Histamine ELISA Test Kit has the capacity for 96 determinations or testing of 42 samples in duplicate (assuming 12 wells for standards). Return any unused microwells to the foil bag and reseal them in the original package with the desiccant provided. Store the kit at 2-8°C*. The shelf life is 12 months when the kit is properly stored.
