Beta-Hydroxybutyrate Assay Kit for the Quantitative Determination of Beta-Hydroxybutyrate in Serum

The MaxDiscovery™ β-Hydroxybutyrate Assay Kit is a plate-based colorimetric enzymatic assay for the quantitative determination of beta-hydroxybutyrate in serum samples from rodents or other mammals. The kit uses a spectrophotometric assay to detect beta-hydroxybutyrate directly from serum samples, enabling researchers to detect beta-hydroxybutyrate levels in animal serum and other tissue matrices such as urine and ascites.

The MaxDiscovery β-Hydroxybutyrate Assay Kit is a simple, accurate and automation-compatible method for measuring beta-hydroxybutyrate (BHBA) levels in serum samples. The kit uses a coupled enzyme reaction to detect BHBA as follows. First the BHBA is converted (oxidized) to acetoacetate by the enzyme BHBA dehydrogenase (BHBADH); in the process NAD+ is reduced to NADH. In the second step, the NADH product is used by the enzyme diaphorase to reduced a colorless tetrazolium salt, INT, to a brightly colored (red) formazan product, which absorbs light at 505 nm. Therefore the observed color change at 505 nm is directly proportional to the β-Hydroxybutyrate concentration in the serum sample.

The kit contains sufficient materials to rapidly test 42 serum samples in duplicate. It also comes with a control solution containing a β-Hydroxybutyrate standard (1 mM) which can be used to calibrate the assay.

β-Hydroxybutyrate, also named 3-hydroxybutyrate, is a ketone body primarily synthesized in the liver from the oxidation of fatty acids. Blood β-Hydroxybutyrate (blood ketone) values provide information about impending or present ketoacidosis and ketotic states due to diabetes or other conditions. The measurement of β-Hydroxybutyrate is essential for determining the severity of diabetic coma in diabetics. The insulin requirements are also based on the blood levels of β-Hydroxybutyrate.
