Acid Phosphatase Assay for the Determination of Acid Phosphatase in Serum

The MaxDiscovery™ Acid Phosphatase Assay Kit is a novel, plate-based colorimetric enzymatic assay for the determination of the acid phosphatase enzyme in 30 µL of serum from rodents or other mammals. Acid phosphatase is an enzyme expressed in a variety of tissues, such as prostate and bone, which removes phosphate groups from phosphocompounds such as phosphotyrosine groups. The enzyme is an important marker for a number of disease states including prostate cancer, hairy cell leukemia and bone diseases. These conditions result in increased levels of acid phosphatase in blood. Therefore, monitoring serum levels of acid phosphatase enzyme has become a routine and fundamental means to detect and monitor various disease states including enlarged prostate, osteoporosis and other bone diseases.

Serum samples can contain two different types of acid phosphatase enzyme which differ in their sensitivity to the enzyme inhibitor tartrate. Tartrate-sensitive acid phosphatase is derived from the prostate and is a marker for prostate diseases while the tartrate-resistant enzyme is derived from bone and other tissue sources. Tartrate is added to serum samples in order to distinguish levels of prostatic acid phosphatase from those of non-prostatic enzyme. The amount of total acid phosphatase activity inhibited by tartrate is equivalent to the prostrate acid phosphatase while the residual acid phosphatase activity represents the non-prostatic acid phosphatase activity.

Acid Phosphatase Assay Protocol

The MaxDiscovery Acid Phosphatase Assay Kit uses a unique colored reaction for specific visible detection of acid phosphatase activity during a 10 minute time interval. In this method, naphthylphosphate is cleaved by the enzyme to form naphthol and phosphate. The naphthol product then reacts with Fast TR reagent included the Reagent Mix to form a colored product. The concentration of acid phosphatase in each sample is determined by measuring the increase in absorbance at 405 nm in a 5 minute time interval. To distinguish prostatic from nonprostatic acid phosphatase (TRAP) enzyme, sodium tartrate is added to the reaction to selectively inhibit the prostatic acid phosphatase. A Naphthol Control, included in the kit, can be used to construct a standard curve to calibrate the assay and confirm assay linearity.
