HER-2 is recommended for detection of Neu of human origin by flow cytometry (1:10 dilution of a 50 µg/mL stock to 100,000 SKBR3 cells), by western blot (starting dilution to be determined by user, dilution range 1:10-1:200), and immunofluorescence (starting dilution to be determined by user, dilution range 1:10-1:200).
HER-2, Human Epidermal Growth Factor Receptor 2, also known as erbB2, belongs to the erbB family of receptor tyrosine kinase proteins. This family also includes epidermal growth factor receptor (EGFR), HER3 (erbB3) and HER4 (erbB4). These cell membrane receptors are composed of an extracellular ligand-binding domain, a transmembrane lipophilic domain, and an intracellular tyrosine kinase domain. Amplification and over expression of Neu occurs in approximately 20-30% of breast carcinomas.
