MaxSignal™ Crystal Violet/LCV ELISA Test Kit (patent-pending) is designed for routine screening for Crystal Violet/LeucoCrystal Violet in fish, shrimp, fish/shrimp feed or water samples of Fish farms or fishponds.

Crystal Violet has been widely used all over the world in the fish farming industry as a fungicide, ectoparasiticide and disinfectant. However, it has been shown recently that Crystal Violet is highly cytotoxic to mammalian cells and has been implicated in the induction of thyroid and liver tumors. This has led to a prohibition of Crystal Violet for the treatment of fish and shrimp used for food production.

To prevent Crystal Violet residues from entering the food chain, both producers and government surveillance agencies need technologies and methods that provide rapid, accurate, and reliable results at the required sensitivity. Currently, Crystal Violet residues are determined by either HPLC or LC-MS method with extremely expensive and time-consuming preparatory procedures. The MaxSignal™ Crystal Violet/LCV ELISA Test Kit provides a simple, rapid, sensitive and cost-effective enzyme immunoassay (ELISA) screening method, which enables government agencies, seafood processors, as well as quality assurance organizations, to detect Crystal Violet to as low as 0.1 ng/g or ppb level in fish and shrimp matrices. The unique features of the kit are:

1. High recovery (80 - 95%), and rapid extraction method for fish/shrimp or water samples.

2. Converting leucocrystal Violet (LCV) to Crystal Violet before detection. Chromatic Crystal Violet can be rapidly bioconverted into colorless LCV in fish (~80%). The special Oxidant Solution in the kit can convert LCV back to the Crystal Violet form which can be detected by the kit.

3. Providing a 96-well high throughput quantitative detection method. The detection limit is 0.1 ng/g or ppb for fish/shrimp samples.

4. The prepared samples can be directly used for liquid chromatography confirmation.

5. High reproducibility.

Procedure Overview

The method is based on a competitive colorimetric ELISA assay. The Crystal Violet (CV) antibody has been coated in the plate wells. During the analysis, sample is added along with the CV-horseradish peroxidase (CV-HRP) conjugate. If the CV residue is present in the sample, it will compete for the CV antibody, thereby preventing the CV-HRP from binding to the antibody attached to the well. The resulting color intensity, after addition of the HRP substrate (TMB), has an inverse relationship with the CV residue concentration in the sample.
