Product Description

This product is the first commercially successful ELISA kit for detecting Malachite Green/Leucomalachite Green (LMG) in fish, shrimp, fish/shrimp feed or water samples of fish farms or fishponds. Malachite Green (Basic Green 4) has been widely used all over the world in the fish farming industry as a fungicide, ectoparasiticide and disinfectant. However, it has been shown recently that Malachite Green is highly cytotoxic to mammalian cells and also acts as a liver tumor-enhancing agent. This has led to a prohibition of Malachite Green for the treatment of fish and shrimp used for food production.

To prevent Malachite Green residues from entering the food chain, both producers and government surveillance agencies need technologies and methods that provide rapid, accurate, and reliable results at the required sensitivity. Currently, Malachite Green residues are determined by either HPLC or LC-MS method with extremely expensive and time-consuming preparatory procedures. The MaxSignal™ Malachite Green/LMG ELISA Test Kit provides a simple, rapid, sensitive and cost-effective enzyme immunoassay (ELISA) screening method, which enables government agencies, seafood processors, as well as quality assurance organizations, to detect Malachite Green to as low as 0.1 ng/g or ppb level in fish and shrimp matrices. The unique features of the kit are:

1. High recovery (80 - 95%), and rapid extraction method for fish/shrimp or water samples.

2. Chemical conversion of leucomalachite Green (LMG) to Malachite Green before detection. Chromatic Malachite Green can be rapidly bioconverted into colorless LMG in fish (~80%). The special Oxidant Solution in the kit can convert LMG back to the Malachite Green form which can be detected by the kit.

3. Providing a 96-well high throughput quantitative detection method. The detection limit is 0.1 ng/g or ppb for fish/shrimp samples.

4. The prepared samples can be directly used for liquid chromatography confirmation.

5. High reproducibility.

Procedure Overview

The method is based on a competitive colorimetric ELISA assay. The Malachite Green (MG) antibody has been coated in the plate wells. During the analysis, sample is added along with the MG-horseradish peroxidase (MG-HRP) conjugate. If the MG residue is present in the sample, it will compete for the MG antibody, thereby preventing the MG-HRP from binding to the antibody attached to the well. The resulting color intensity, after addition of the HRP substrate (TMB), has an inverse relationship with the MG residue concentration in the sample.
