Monoclonal Antibody Services

 

BIOO Scientific has developed a unique 5-week Monoclonal Antibody Production Service (patent-pending) to produce custom monoclonal antibodies against peptides and proteins. Using BIOO Scientifics’ cutting edge, patent-pending technology, we are able to deliver a tissue culture supernatant containing specific monoclonal antibody to our customers for assessment within 5 weeks after receiving the antigen, either peptide or protein. BIOO Scientifics’ antibody products and services are complete from antigen design, antigen production, antigen purification, antigen conjugation, host immunization, bleed testing, hybridoma production, and hybridoma screening to antibody purification and characterization and ascites fluid preparation. BIOO is a leader in the production of antipeptide and antiprotein antibodies. To enhance the solubility, purity, and antigenicity of a selected peptide, we express peptides with our unique vector instead of conjugating the target with a carrier protein. Immunization with our prepared peptide can produce an antibody with higher titer and specificity than with conventional methods. Using our exclusive method we can generate an effective antibody where other systems fail.

The Stages of Monoclonal Antibody Product at BIOO Scientific:

  • Stage I: Immunization:  Five virus antibody-free BALB/c mice will be immunized with the customer’s antigen.  Peptide, protein or small molecules can be used.  A minimum of 15 mg of peptide or 5 mg of protein is needed for immunization and subsequent screening.  BIOO will charge customers for the extra cost of antigen production/conjugations, if needed.  The mice will be housed in an isolator during the immunization period.  Five mice will be immunized in order to increase the chance of an immunogenic reaction, but only one mouse will be used for fusion.  An additional charge will be invoiced if customers want to use two mice for the fusion.
  • Stage II:  Fusion and Screening:  The popliteal node (if the customer supplies either a peptide or a protein) or spleen (if the customer supplies a small molecule) from the immunized mouse will be harvested and all spleen/node cells will be fused with a myeloma cell line, P3.  All 96 well panels will be screened by ELISA for antibody positive wells.  About 10 positive wells from the ELISA assay will be selected to be subcloned and frozen for backup.
  • Stage III:  Cloning
    • First Cloning: A maximum of 10 positive wells selected will be cloned in 96-well plates. Single colonies will be tested by ELISA and selected positive clones will be expanded for the next cycle of cloning.  Hybridoma cells from these positive clones will be frozen as backup.
    • Final Cloning: Positive clones from the first cloning cycle will be selected for the second cloning cycle.  Wells containing single colonies will be tested by ELISA and positive clones representing the 3 best original clones will be expanded.  Several cycles of cloning may be required in some hybridoma cells in order to obtain true and stable clones.  Cells from these positive clones will be frozen as necessary for backup.
    • Frozen stock and ascites:  The maximal 3 clones selected will be expanded and a small stock of each clone will be frozen as established hybridoma cell lines. The supernatant and/or vials of cells will be shipped to the customer for evaluation.  In addition, one BALB/c mouse will be inoculated with one selected clone of customer choice for ascites production. The ascites fluid will be prepared and shipped to the customer.  BIOO provides affinity purification of ascites fluid or tissue culture supernatant for an additional charge.

CRITERIA OF SUCCESS

  • Bioo Scientific Corp. will make every effort to reduce the production turnaround time. Delays may be incurred if the antigen is poorly immunogenic or produces unstable clones.
  • The success of a project is dependent upon the material used, supplied by either the customer or by our supplier. Some or all animals may fail to give a response to certain antigens. Please note that Bioo Scientific does not guarantee that a given antigen designed by BIOO or provided by customer will elicit an immune response.

PROPRIETARY

  • After the project is completed, Bioo Scientific Corp. will transfer the ownership of the monoclonal antibodies (all the cell lines and intellectual property) to the customer.
